Boston should be included it is the largest financial center in the USA outside of NYC and Chicago and ranks number 6 world wide. It also has the largest bio-medical cluster in the country and is the largest center of university research in the world.

Correct....DC may not have the population but it does have a signifigance out of porportion to its size. I say SF Bay Area due to the population puts it in a sort of big-city status, aside from the obvious intellectual/cultural/technological signfigance.
I''d might add Boston, due to the presence of a lot of tech industry, Harvard, MIT, Tufts, Brandies: an intellectual center.
