I don't mean in demographics, but in overall influence in the culture and architecture of the city? There are many contenders for the title.

In culture, I would have to say Miami. The cuban population is just such a huge part of that city, that it affects the city's culture. It's probably one of the handful of cities in the US where speaking spanish is more important than speaking english in many industries.

In architecture, the sunbelt cities and other cities along the mexican border, are probably most influenced by latin America. Of the cities that I've been to, San Jose was the most influenced by latin american architecture.

Miami without question. I say this as an Angeleno who has also lived in San Antonio for a short, brutal time. And outside of touristy city centers, the architecture argument is not important - if it was, St. Augustine, Florida might be on the list, along with their 17 hispanic residents.

This is it, perfectly.

I would pick Miami if we're only picking one, but the tier system above is perfect.

What does Spanish architecture from Spain have to do with Latin America?

I would switch your tier 3 and 4 with each other. Phoenix, San Diego, San Antonio etc. feel FAR more "Latin" influenced than DC or Boston.
