Probably Jackson, MS. In fact the state as a whole has a very bad and unforgettable reputation as far as racial segregation. I remember hearing about segregated proms in some town there within this decade.

Oh you said MAJOR cities, then in that case perhaps Birmingham or Atlanta.
All the major cities in the south are segregated, Houston, Dallas, Atlanta, New Orleans.. Legacy of the deep south past..
One thing I have noticed at least in Atlanta though is that the segregation is mainly a black and white thing.. Asians and Hispanics are not that segregated compared to African Americans..
The funny part about all these Major Cities and out of all the Cities I have been to in the south, Nashville seems to be that one City that's the least segregated, IMO. All races seem to mix well in the City and the population seem to shame racism Hard there!!

Yes, because those 3 other states aren’t viewed as uniformly being Southern like Virginia usually is. That’s all... Kentucky out of the other 3, is viewed as being Southern in comparison though.

I don't think any if these states, including Virginia, is thought if as uniformly southern. This new thing with not viewing West Virginia as southern is enlightening, to say the least. And anecdotally I think Kentucky is thought of as being most southern of the group...
Regardless, they all are southern. Maryland being the least so, each of the four having culturally blended regions that aren't traditionally southern, or even thought of as southern in the modern sense. Not a single one of the four is devoid of southern culture and all four have regions that are unquestionably southern. Each of the four is easily more culturally southern than the states to their norths. Each of the four is easily distinguishable from from traditional Northeast and Midwest, nay, traditional "northern" culture, and each of the four is also heavily influenced culturally by the Northeast and/or Midwest, each of the four having regions that are blended into or with those regions...

I was thinking the same thing. The map cuts off a large portion of the Southern US, but oddly enough includes Texas and Oklahima, so it is hard to answer the OP's question.
