I knew Calgary and Denver were comparable in many respects, and Calgary is the most conservative of the large Canadian cities. But Denver, however, I do not feel it's the most conservative large city in the US.
For some reason I felt that Calgary was more conservative than Denver but I am not convinced either way. Perhaps my bias comes from the impression that liberalism vs. conservatism, in the Canadian context, is mostly framed in terms of economic issues. I might harbor a whole bunch of misconceptions for what I know.

Denver isn't conservative at all. It's actually one of the more progressive cities in the country. Parts of Colorado are conservative but not the Denver metro area.
As someone who lived very recently in Denver, I would say it is quite socially progressive, even liberal. Boulder, to the North (but still part of the MSA) is very liberal.
However, economically and even in terms of how people in the city view the role of government, I would say Denver is fairly moderate, which is to say, I think it's more conservative about many things than people initially think it is. There is still a big part of the city's economy that is dependent on energy, specifically O&G and mining.
That's just the impression I got while living there, anyway. I loved the balance of people's views and found it really suited me as someone who is socially liberal, but fiscally conservative.
