Gotta be I-80. Goes through the Northeast, Midwest, Mountain West, and to the west coast. In terms of going through the most regions, that's gotta be up there (all though it doesn't go through the south).

I don't know how I overlooked I-80. Maybe a moderator can add it to the poll.
Yeah I vote 80. 95 might have more significant cities along it, but 80 really bifurcates the nation more while also having some significant cities. It's kinda how I think of Burnside as being Portland's main street, even though you can find streets that pass more centrally through the downtown.
Another vote for 80- SF, Oakland, Sacramento, Reno, SLC, Omaha, Des Moines, Chicago, Cleveland suburbs, NYC (ok, it ends in NJ just before the George Washington Bridge).
It goes from the Bay Area Area, through the Sierras, across some deserts, over the Rockies, across the Great Plains, near the Great Lakes, and across the Appalachians in Pennsylvania on the way to NYC
Yet another vote for 80. Was my first thought when I read the post title. Ditto on everything that's been said about it passing through so many regions and significant cities.
