I do think this really is between Philly and Boston, I lean to Philly really as the Declaration and Constitution being implemented there is the differentiator to me which to me are just hard to deny as the foundational structure/seminal documents of the US experience.
History is by means solely limited to these two but both have what to me is the richest history of major events that formed all US history to follow.
Baltimore is very important in US history. I'd even argue that it rivals Philly and Boston. I guess the only difference is that the other two are slightly older.

New Orleans created the Soul of America and created American pop culture before Jazz America was trying to be Europe.
Because New Orleans did not start as a English Colonial city or was ever ruled by the British. It reflected a culture that was closer link to Latin American Havana Cuba or Salvador Brazil. Then Boston or Philly early on.
Jazz is America first true pop music art form, it completely change American society and morality. None of the music forms from Rock, R&B, Hip hop, to Edm would happen if wasn't for New Orleans birthing Jazz. That's why I said New Orleans is extremely important in American cultural history, because with out we have alternative history.
