After updating windows 10 with the April 2018 update I am getting this error when launching one program.
.NET Framework Initialization Error
Please set registry key HKLM\Software\Microsoft\.NETFramework\InstallRoot to point to the .NET Framework install location
After extensive searches online and attempting all the fixes it seems as if I have to do a repair install of windows (hopefully) or a clean install worst case if I ever want to use that program again.
I have uninstalled and installed the program throwing the error and this did not help.
In the start menu under Windows System right click command prompt and select "Run as administrator". Enter this command:
sfc /scannow
That checks and will repair windows files and AFAIK it works for NET Framework installations. It may or may not fix it.

I appreciate the suggestion. I know just enough to be dangerous when it comes to messing with the registry.
It really is straightforward. The most important thing is to Click on the File menu then Export before doing anything. That way if it gets wonky you import that saved file and the changes are gone.

Windows key+R then type regedit
