Make sure ALL the volume controls are on. Most PCs have multiple controls, and a zero volume on any of them shuts up your speakers/headphones. Obviously, the volume control in the tray area (looks like a little speaker, in the lower left area next to the timeclock, usually). But you should also right-click the volume control and then click the "Open Volume Control" to see some other controls that might be giving you a problem. And if you're running some program that plays sounds, check that it doesn't have it's own volume control somewhere on its screen.
Ah, but if the headphones are working while the speakers are not, it probably is a problem in the headphone jack itself.
