recently I've been having some weird, random volume problems with my tube guitar amp even after retubing it. Some one told me when that happens to unplug the amp and keep it unplugged over night, something about an electrical build up in the amp. Has anyone ever heard of this, is there any science behind this?

No. That is just plain stupid. Any electric "buildup" would be from caps not discharging, and a fix wouldn't happen that way.

Random volume problems is pretty non-specific. If the amp is a push-pull (paired 6L6s, etc.) then a dropout of one of the pair could cause that. Exact diagnosis would mean tracing out the circuit. First guess would be a cold solder joint, bad resistor or cap.
