Interlocken in a business park in Broomfield that contains a lot of high tech companies such as Sun Microsystems, Level 3, and a bunch of smaller employers.
Homes in that square foot range for that price range might be a bit hard to come by, but I do see some homes listed in both the Broadlands and Aspen Creek (neighborhoods in Broomfield) in the low 5,000 sq foot range in the mid 600s. I would have suggested my neighborhood, Bradburn Village in Westminster because we're packed with kids, super social, a great location for techies (we border Broomfield), and a walkable, new urbanist neighborhood (we can walk to lots of stores and restaurants) but we don't have many homes above the 4,000 sq foot range (although there is one model that is 4200 that with a finished basement would be 5200, but it would be in the 700s for sure).
Good luck with house hunting!

I would have to agree with Steve, your square footage and price range don't seem to match, why the need for such a large house?... I agree that Broomfield would be your best bet at finding a job and a home that will fit your needs.

So if there was a 15-30 minute "light rail" option, this would probably be just fine. Given this, are there any nice neighborhoods that would meet most of the items on our "wish list" that would have a good light rail option to any of the best tech employment centers?
One of the most important requirement is a large home 5500-7000 sq/ft for between $400,00 and $650,000 so any neighborhood needs to have homes in this size/price range.
Thanks in advance for any advice you can provide!

Interlocken is simply the name of the suburban office park along US-36. It's like the "Denver Tech Center" in the south metro. I'm not sure where the boundaries are, but it's in the vicinity of the flatirons shopping mall and storagetek drive, including the giant Sun Microsystems campus, and all the offices on the north side of the Jeffco Airport (I believe it's now called the Rocky Mountain Airport).
I think your expectations for square footage is too high for your price range. Homes of that size in close-in locations would be well over a milion. If you're willing to live quite far out you might find something at the very top end of your price range. There's just very few homes in that size range at any price, particularly if that's above ground square footage.

What about SE Aurora? It doesn't seem to be all that far from the DTC and there are lots of homes listed in our price range. We need the space for a home based business that requires lots of storage. I'm a bit worried about that the zoning might not allow for this, but we wouldn't have any customers stopping by. We're simply mailing out products. So the fed-ex and UPS might stop by more than usual, and U.S.P.S. will have a daily pick up of a few baskets of orders, but unless someone was really paying attention, I'm not sure how anyone would even know we were running a business. Do you think I should be concerned about this??

Home based businesses are usually no problem, especially if you have no employees in addition to yourselves. In a newer neighborhood, of course, the pesky HOA might give you grief, but I don't think the city will.
SE Aurora is one of the more affordable areas in town, and it looks like there may be one or two homes in your size range that are also in your price range there. The main problem is that in parts you're a somewhat captive audience for the E-470 toll road, which can get expensive. This is less so for the areas closer to I-225, of course.

The city of Louisville (for one) has a number of regulations for home-based businesses. You should check before you rent or buy.

Yeah, but he's looking for "urban estate" square footage on a KB budget. And I have a feeling he'd consider our lots "postage stamps", although I measured and my lot would not fit on an envelope by any means But I love my little yard that takes 10 minutes to mow - and I'm on a corner lot!
