Where is the city center? What landmarks do I see? What do I do? I can get a lot done in 3 days. I am not the type to go and waste time in the hotel. I hear Bairro Alto is great for nightlife.
Take a half day tour and see the major city sights. If you like museums the Gulbenkian is terrific. Enjoy the beautiful public squares--Rossio, Praca do Comercio. Sit out at Cafe. Sweeten your diet by eating pasteis de nata. Walk and get lost in Alfama on the way up to Castelo S.. Jorge for great views, sample the thriving night life in Bairro Alto. Hear fado singers in a nightclub.

In addition to all of these you can take day trips to other places outside Lisbon such as Cascais/Estoril and Sintra.
If you're looking for more modern architecture next to the river head to Parque das Nações.
Do a free walking tour (just make sure to tip well)!
Go to a restaurant called Super Mario.. very authentic, local feel to it.
