Turnpike north to I-10, 10 west to Phoenix, AZ 60 to Wickenburg, AZ 93 to Boulder City (get to cross the Hoover Dam), I-515 into Vegas.

You gotta take I-75 in Ocala which will connect you with I-10 in Lake City FL

You can also take I-95 to I-10 in Jacksonville but that would much longer(probably 1-2 hours) but you won't pay the Turnpike tolls which I think are around $20 total.

I have made that route from Miami to Mobile, AL but I can tell you the roads are very good. It's only inconvenient if you get heavy rain.

Depends what you mean by best
If you're in no hurry then motoring around the Panhandle via the scenic 98 route is an absolute delight.The roads are deserted in most places too.
I'd recommend overnighting in Apalachicola.
