Smith River, CA (NW Corner) to Andrade, CA (SE Corner) is about 1,000 miles and probably a little under 20 hours away (according to Mapquest).

1,004 miles and 16:26 according to Google Maps - New Leader!!! Can anyone do better?

Littlefield AZ to Douglas AZ can be 11 hours and 672 miles, but you have to go out of state to get there.

While it's not an extreme point to extreme point oddity since all of Ohio's extreme points are near interstate highways and Ohio doesn't really have an "extreme southeast point", but driving along the Ohio River the entire way from East Liverpool to Cincinnati takes over 8 and a half hours.

Just to put in a vote for NC, From Ducktown in the Smokies to Hatteras Island at the shore is 519 miles and takes 8 hours 32 minutes. Raleigh is just about midway and so is located just where a State capital should be.

There's also a town simply called "Duck" on the Outter Banks that's likely even farther from Ducktown than Hatteras Island is.
