Houma/Thibodaux-Baton Rouge-Natchez-Monroe-Little Rock is a needed route.
Augusta-Macon-Columbus-Montgomery-Hattiesburg-Natchez-Alexandria-Waco is the proposed I-14.
In northern New England and northern New York, there is no east-west interstate. So when I went to northern Maine, I had to go south first and then north again; either that or take a slower back road. Clearly the presence of mountains limits where such a highway can be built.
Useless highway? They're building one (albeit a short one) in Prince Edward Island. I took the old route on the first Saturday in August and had no problems getting anywhere. The islanders didn't want it and I see why.
Most useless: Any "beltway" or "loop" interstates. This includes Washington DC's Capital Beltway (I-495), San Antonio's I-410, Atlanta's I-285, Rochester's Inner Loop (part of I-490), Harrisburg's beltway (I-81 and I-83), Columbus' Outerbelt (I-270), Kansas City's I-435 and many, many others.
Most needed: None. We have more than enough highway infrastructure in the United States as long as we maintain it.

Both loops in Houston are needed, now I'm not so sure about the new Grand Parkway being built.
You have to be nuts if you think the freeway infrastructure of the 40s-50s is supposed to last us as the country continues to grow. My metro area alone is suffocating under the lack of freeways, not to mention this entire state.
All interstates to the Canadian border are now pretty useless, since nobody is willing to endure the ordeal of trying to cross the border. Particularly the I-75 from the Mackinac Bridge to Sault Ste. Marie. Anybody who is driving on that road is a suspected terrorist, and often pulled over and questioned, especially if you pull off the highway "for no apparent reason". Sault Ste. Marie is a town of 14,000 with a 50-mile interstate highway connecting it to the rest of the world. With overpasses and cloverleafs, to avoid traffic snarls like this:

Considering they are still our #1 trading partner, this is flat out false and dead wrong. Also go to the Peace Arch crossing on I-5 on the WA/BC border on a Friday afternoon and then tell me how useless US/Canadian Interstate highway border crossings are.
