I am aware of the subjective nature of this question, but I'm curious to see what some opinions are. I've seen lists generated online but I'd like to hear some of your thoughts on this.
For me, Houston. I have a lot of family in Texas. Some of them love Houston, some of them hate it. After trying to live there twice, it just isn't for me. Most people seem like they are in such a hurry. Road rage is severe. Crime is high. Some people are friendly, but many more are downright rude. It is flat, ugly, hot, humid, smoggy, and lacking in character. Anyway, the people I know who don't like Houston have left. But I seem to encounter a lot of miserable people every time I visit the city. I know there are those who love it (some of my relatives included), so please don't go bashing my opinion. It is just that. But since it has been asked, that is my take. I have no good answer why so many Houstonians seem miserable. Perhaps if all the haters moved away, the place would seem a bit more hospitable?
For me, its gotta be NYC. The stress levels, the traffic 24/7, the high cost of living, and the frantic pace all combine to make people pretty miserable. There's good things to NYC also, to be sure, but overall its a really tough place for middle income people to live.
Los Angeles has got to be one: urban sprawl, road rage, horrendous traffic congestion, 40% of the population cannot properly speak/write in English, high unemployment rates, lots of gangs...

Edit: I actually stopped reading once you said 40% of people can't properly write or speak English.
