I've seen a lot of posts mentioning that City X's downtown is dead after 5 PM as if that's a sign that nothing goes on in that town. In my experience, downtown is very rarely where you want to be at night in any city. What city's CBD is actually its primary nightlife district? Houston, maybe?

You are way off the mark.
Houston has no primary entertainment district.
To me every time I visit the hot spot changes.
Richmond used to be the hot spot in the late 90s.
In the early 2000s it was main.
Then it was downtown.
More recently it was Washington ave.
I have not gone out while in Houston in the last couple of years but the it spot might have already moved from Washington Ave.
KC, Cleveland, Memphis, Omaha, Austin, Nashville, San Antonio, San Diego and maybe even New Orleans and Chicago all have their primary or at least major nightlife districts in their downtowns or "greater downtowns", though not always in the CBD.

Austin's 6th St and the Warehouse district are CBD adjacent, as is the French Quarter. River North has a fair amount of Chicago's nightlife, but it's outside of the loop, and Wrigleyville is probably the biggest nightlife destination, followed by River West/Fulton Market/Randolph and Bucktown/Wicker Park.
I think that this is generally the case with mid sized and smaller cities/college towns. Even some of these places will have other neighborhoods/districts that have nightlife as well. For instance, here are a couple of Downtown districts near me: Downtown Syracuse, NY Restaurants, Bars, and Shopping

Definitely not Houston. Houston has two great whiskey bars downtown, but its hardly a nightlife district. The only nightlife districts Houston really has are Midtown and Washington Ave.
Seattle spreads nightlife to numerous districts -- Pioneer Square, First Ave & the Market, Belltown, Pike/Pine, Lower Queen Anne, Fremont, Ballard, and others are close-in examples. A handful of districts are on the fringes of the CBD.
