I'm thinking of towns where a large, high-end hotel has been their traditional centerpiece.

Offhand I can think of:

White Sulphur Springs, WV (The Greenbriar)
Hot Springs, VA (The Homestead)
Colorado Springs, CO (The Broadmoor)
Mackinac Island, MI (The Grand)
Asheville, NC (Grove Park Inn)
Estes Park, CO (The Stanley)

Any others? I'm not really looking for this type of hotel in a major metro, just in relatively small communities.

The Mt Washington Hotel in Carroll NH is a classic.

The Penn Wells in Wellsboro, PA is prominent, the place where one meets, etc. https://pennwells.com/ It's not high-end in the sense of The Greenbrier, though.

Bedford Springs in Bedford, PA is a high-end spa hotel more in the sense of The Greenbrier and The Homestead. https://www.omnihotels.com/hotels/bedford-springs However, it's not directly in town, but a couple of miles out.

Quebec City, QC might be considered small by some, and Best Luxury Hotel in Old Quebec City - Fairmont Le Château Frontenac is an undoubted centerpiece.

Centerpiece hotels are not uncommon in small towns in PA. High-end? Not exactly.

For Canada I would say the Banff Springs Hotel in Banff Alberta, just stunning! I actually worked there in the late '80s.
