Can you cool a sustainable building without using natural ventilation? Is natural ventilation the MOST efficient way of cooling a building?

Depends on the climate in the area. In Florida, no. Too much humidity,

Use a fan to pull in cooler night time air and keep the place closed up during the hot part of the day. If you are in a really hot climate try and use natural ventillation as much as possible. That is live outdoors behind insect screens.
