Hi how goes it?
My Wife and I plan to buy a home in 8 to 10 months with our Mother-In-Law. Our MIL wants to live in a small town where you walk into a Post Office they know your name she told us.
Wondering thoughts on Kernersville, Jamestown, Thomasville, Colfax, Clemmons, and Trinity???

Out of the places you list, maybe (?) Thomasville or Trinity would match that request.
The reason I say this is because while Kernersville, Jamestown, Colfax, and Clemmons may have a certain quaintness (Colfax is more of an area than a real town though), they're growing and busier areas closer to the core of the Triad. Those areas will likely continue to feel busier and denser with people who are not from the area moving in and out more, so I don't know that this will fit the bill in terms of that overall local vibe. She can get to know people anywhere in the Triad, and maybe have that vibe, but I just don't know that those areas are going to give her the feel she's looking for. While Trinity and Thomasville might be growing a little, they feel much closer to how they felt 20-30 years ago than the other places on your list. They're fairly friendly, I suppose, just your typical semi-rural, blue collar working-class Southern communities. She may love this or not, but Trinity and Thomasville are both politically "red" areas.
To find the vibe I think you're talking about, your M-I-L would be looking at some much smaller towns in the outlying areas of the Triad, particularly outside Guilford and Forsyth counties. And then, some of the locals can be friendly and some can be kind of standoffish too, especially when it comes to newcomers, so it's not entirely Mayberry and Cheers.

Looking at about $300K 3 to 3 bed house and 2 baths with some land hopefully. Trying to be 20 miles or less out from Winston-Salem.

Walk to post office? Most of the places recommended you will have to drive as there will be only one. lol, but if you mean you want the folks to be welcoming, friendly and warm, then you may want to be not too far 'out' where homes are farther apart and neighborly chats are more difficult. Where are you coming from, if I may ask? Some areas around WS and the Triad are more welcoming of 'yankees' than others. Yup, still a thing. There are pocket neighborhoods all over that may give you what you want, if it is nice 'sidewalk' neighbors that cookout and have that 'drop by for a drink' attitude.
So, it would be helpful for you to be really specific as each town has pockets that could meet your criteria. How much land? Close to neighbors? Do you want restaurants? Cafes? Pubs? Jamestown is a great little town and right off their 'main' street are homes with land as well as neighborhoods with larger yards. And as Jowel said, Clemmons is another nice area.

You might look at Lewisville, too, but all small towns around here will be relatively car-dependent.

Walkertown (3 miles from Winston) , Stokesdale (9), Reidsville (19), all along us-158
Reidsville is a city, but is still small slow-paced and has parts that are rural
