This is a fun subject and all, but kind of irrelevant with Six Flags in Atlanta and Carowinds in Charlotte. It would never happen.
It could happen someday, but I doubt "the Upstate" will have much of a voice if it ever does happen, unless a signifiant incentives package is offered by a conglomeration of Upstate municipal organizations.
Something similar to Family Kingdom in Myrtle Beach could be interesting. It is much smaller than your typical amusement park.
At one point, the Greenville Zoo was said to be thinking of moving to a larger space one day. So if the Zoo would move, the land there could be used for the Family Kingdom size of park. It would also bring a lot more people to the downtown area. (I realize parking is an issue, plus dozens of other things, but I'm just playing along...)

I was thinking of a smaller scale as well. Family Kingdom has a decent amount of rides but is not huge like the other parks mentioned. It would be good for the kids and teens. Maybe have a wristband for unlimited rides or buy individual tickets. I don't think a water park would be a bad idea either. I know there are some around for the little ones but a bigger one with water slides, lazy river, tube rides and wave pool.

I just don't see there being enough of a market for a significant theme or amusement park to make it will all the competition in the regional neighborhood (which could also include Dollywood).
Are we forgetting about the failed park (Hard Rock) in Myrtle Beach?
With how badly that thing failed, it will be a LONG time before the state ever sees a new one.
Hard Rock failed for numerous reasons, such as ticket price @ 50 bucks (ouch), location, poor management and a soft economy when it opened (bad timing).
A smaller niche park, like Family Kingdom, may do well after the economy gets moving again.
