There are revolvers made which have interchangeable cylinders - 22 WMR (22 mag) and 22LR.
You can use the significantly cheaper 22LR to practice and load up the 22 WMR when it counts.
We have an 8-shot 22 WMR revolver loaded up with Speer Gold Dots. It's not the primary go-to piece if someone kicks in the front door at 2am, but it's another option if we ever needed it. It's fun to shoot at the range and I'm sure it'll stop a bad guy.
Not saying .22 is the ideal defense weapon, but it's better than having nothing.
I would agree with the suggestions for a revolver. The first factor is whether you're going to be dedicated enough to learn, practice, and maintain proficiency in the more complex "manual of arms" (technical operation) of the semi-auto compared to the revolver. If not, all the other factors of choice--pros, cons, mitigations, risks, benefits--are irrelevant.

Wouldn't 22WMR be too great a risk of overpenetration if used for HD? Other than that, it sounds pretty great.

I'm going to see if I can get the wife to like revolvers. She doesn't care for the look so much; she likes the semiautos, which we've both shot before in 9mm. She handled it OK, but it wasn't 3AM.

No recoil to speak of, huge capacity, almost as quiet as my old Red Ryder, cheaper than last year's iPhone, and what? A 9-round revolver?! That just sounds FUN.
The one thing I worry about: If a bad guy breaks in and I shoot that entire cylinder at him and am sued by his dirtbag family, won't a jury think I went full rampaging Eastwood/Heston on the dead perp? "The defendant shot the deceased victim NINE times, ladies and gentlemen!"
Of course, if dead dirtbag was in my house uninvited at 2AM, I'd have a pretty solid defense.
But is there a risk of overpenetration with .22LR? That to me is a serious consideration--and another point in favor of the modern-Minie-ball/Celtic hurling stone that is the .45acp round. (Plus it looks like .45 is not that expensive these days--just a few cents more per round.)
You can take someone down with one shot. And .22LR does more bodily damage than any other gun you have mentioned. A .22 moves through a body like no other projectile. Just sayin. A .22 is a nice, fine little handgun that is cheap to shoot and easy for your wife to handle.
I sleep with a 12 gauge shotgun, but I am not typical. I can hande a .40 cal, but most women cannot handle the recoil.

If someone breaks into my house, and I have a .22LR or .380, I imagine I am going to shoot them until the chamber is empty. Actually, stress might lead me to do that no matter the round.
I wouldn't consider a 22 for protection at all IMHO. Its all fine and dandy to say you can hit a small grouping, but in a stressful situation, you really don't have a lot of time to take to aim. You want something that will have an impact, and not just **** someone off.
My wife has a Sig 938 (9mm). While its a little small for my hands, its a nice gun, and shoots well. It fits her hands better than my M&P Shield 40, which is my usual carry.

Have you taken a handgun class? Does your state have requirements for training?
Have you ever hunted? Shot anything? Your post is concerning.
You'd be better off with a 380 or a 9mm instead of a 22. I like 22s. I still have the first 22 rifle given to me as a first as Christmas present when I was 10.
