Great Question! As far as the choices you named, I would go with Rome vs. Carthage for sheer ferocity and the amount pitched battles. As far as longevity, the Israeli-Palestinian feud has got to take the cake--take the falaffel?--but I would have to rule this particular squabble out as being the Greatest simply because it has always been pretty one-sided, with Israel perenially kicking Arab butt! (Remember that we give Israel more money and arms than we do any other country.)
So my humble entry can be found right here at home: The North vs. the South in the Americsn Civil War. Or, as the south STILL calls it: "The War of Northern Aggression." LOL. This war changed our country's future and the ramification from it still live on today. Over 600,000 Americans died, and if you do the math, you'll find that as a percentage of the population back in 1861, that is a truly staggering number. It still blows me away the ghastly number of casualties that some of those battles posted, like 50,000 in three days at Gettysburgh. Or Grant losing 10,000 men at Shiloh.
And even today there still lives a sort of idealogical and cultural rivalry between American Northernors and Southerners.

I think Germany v USSR was a lot bigger than USA v USSR. Certainly more people killed.
Queensland v NSW in 'State of Origin'
Aus v England in cricket

Cuz he's also a giant and Godzilla only has a shot if his fire hits the target. Close combat, he's toast. Don't believe me? OK how about you and I fight close combat - but your arms have to be tied to your sides all T-Rex like.
Anyway, given the extremely broad premise (which IMO means this should exit the history forum, frankly) - I say it's easily a 2-horse race: liberals vs conservatives or men vs women.
Or Colts vs Pats. nyahh nyahh and lol @ BB.
