How flexible are you on the route? Are you planning on taking as much interstate as possible, or other state highways to see a little more?

It looks like our route brings us in on I-80, north on I-35 then east on US-20. We have about 2 hours to detour for sight seeing.

Wow, 2 hrs isn't very much!
That route, coming from the West, will take you right by the exit for the Living History Farms in Urbandale - just west of the metropolitan Des Moines area, and located where 80 and 35 merge. The LHF can be done in 2 hrs - but just barely! Here's their website so you can think about whether or not this would interest you:

Unfortunately it's probably too far out of your way, but Mount Sterling is single best place in Iowa to visit.

I'd take the I-235 exit (off of I-80) as this will bring you (somewhat) through the heart of the metro. Get off on a downtown exit and check out the Sculpture Park, take drive through downtown to explore a bit, drive east towards Capitol Building (check that out if you have time).
There are a lot of good places to eat downtown depending on what you're interested in trying.
After getting done downtown, you simply hop back on I-235 heading east again and you'll connect again with I-80/I-35.
