Unfortunately, the answer is not downtown properm but very close. I am not sure the store listed above will have what he needs, but I think it may have basic button down shirts. The place mdawg listed is only a 3 block walk from his hotel, so it may be worth a shot. Anyways, thisis why Louisville desperately needs the center city retail project to start soon, and it is the major weakness downtown right now (lack of clothing retail which left downtown in the past 10 years). If interested check the development thread, but this wont help your husband in the next 24 hours.
The closest and easiest place to get a nice, fairly priced men's shirt close to downtown is in GreenTree Mall in Clarksville, IN. There is a nice Dillard's there with men's shirts, and it is about a 7 minute drive or 10 dollar cab fare from his hotel.

If your husband is looking for higher end stores than the basics available across the bridge, he'll have to head to the east side of Louisville, which is currently the clothing retail center of the metro, especially for business. Here he can find higher end stores like Brooks Brothers, Banana Republic, Von Maur, etc. There are also upscale boutiques for higher end shirts:
