This really depends on what you're interested in--architecture, museums, restaurants, clubs????

To Slig's list, I'd add the Crystal Court in the IDS Center--it's basically the heart of downtown (especially during the workday), otherwise there are good museums (Walker Art Center & Mpls Sculpture Garden (w. the Cherry on the Spoon), and Mill City Museum); Farmer's Market on Nicollet Mall on Thursdays and at the Guthrie on Saturdays. Nicollet Mall between 7th Street and 12th Street is an interesting walk--lots of sidewalk cafes.

I was gonna mention some of those things but couldn't go as far as calling them "must sees." The Crystal Court and the Farmer's Market are pretty cool though, I will admit.
