my wife and i are planning on relocating to missouri, i have found a job in jeff city, her in st louis, we know this is quite a distance apart but would like to know if it is possible to live in between and commute, any feedback would be appreciated.

Which part of StL will she be?

Yes it is possible, and quick suggestion would be somewhere near Warrenton, off of I-70.

High Hill would be about the most equidistant, but don't know that much about that particular area, or if that would even be suitable for your needs - rather live in smallish town, rural/country/farm, outskirts, school situation, shopping, etc.?

Please keep in mind that traveling into the city [StL], depending upon time and weather and road conditions can be substantially greater than the ~60 minutes that google/mapquest etc. will give you.

Going towards JC, I haven't traveled on US54 that much, sorry can't help you on that section.

BTW, along route 94 is very scenic areas - wineries, KATY trail, etc.

Default Some ideas for you

The far western suburbs like Warranton and Wentzville offer a wide variety of homes that wouldn't be that terrible a drive to Jefferson City. You could also look out Interstate 44 near Washington or Union. Hermann is nice too.
