If I had to pick just one (because I pretty much collect great movie lines)....

Big Bob, as played by Bruce Dern, in the movie "Smile". He's a used car salesman pounding on the side of a Winnebago that he's trying to sell to a husband and (pregnant) wife.

"Heck of a secure feeling knowing you're sitting on a hundred gallons of gasoline."

From Bladerunner. Roy Batty, played by Rutger Hauer:

I've seen things you people wouldn't believe. Attack ships on fire off the shoulder of Orion. I watched C-beams glitter in the dark near the Tannhauser gate. All those moments will be lost in time, like tears in rain. Time to die.

I've posted these before, possibly even in this thread, but they are worth repeating, as great lines, in great (often not widely seen) movies; Still, they're worth posting again.

Maria; I can explain everything
Kynaston; Why, are you a philosopher?

Maria; Your old tutor did you a disservice Mr. Kynaston. He taught you how to speak, and swoon, and toss your head but he never taught you how to suffer like a woman, or love like a woman. He trapped a man in a woman's form and left you there to die! I always hated you as Desdoma. You never fought! You just died, beautifully. No woman would die like that, no matter how much she loved him. A woman would fight!

Both of these are the beautiful Claire Danes to Billy Crudup in "Stage Beauty".

Johnny; Are you okay little girl?
Christy; Don't "little girl" me! I've been carrying this family on my back for over a year. Ever since Frankie died. He was my brother too. It's not my fault that he's dead. And it's not my fault that I'm still alive.

From "In America".

Anyone who can watch the above exchange and another one later in the movie and not be MOVED, has no soul.

Listen -- strange women lying in ponds distributing swords
is no basis for a system of government. Supreme executive power
derives from a mandate from the masses, not from some farcical
aquatic ceremony.

"Look. I already told you. I deal with the g****mn customers so the engineers don't have to. I have people skills; I am good at dealing with people. Can't you understand that? What the hell is wrong with you people? "
