First and foremost, I would make sure and stay away from the interstate.

If one were to start in...let's say Grand Island? I would take 281 north to O'Neil, then hwy. 20 west to Chadron. From there I would head south through Scottsbluff/Gering down to Kimball then take hwy. 30 back east to Ogalalla then north to Hyannis, then back east through Mullen, Thedford and Broken Bow and back to the starting point. In the end making a very irregularly-shaped circle.

Fully agree with ths statement... and if traveling east towards Omaha from that point, go on Hwy 92 instead of the interstate.
