You need to take the LIRR Montauk Branch out to Mastic-Shirley: http://mta.info/lirr/Timetable/Branch/MontaukBranch.pdf

From there, you can take the S66 or 7E (or a taxi/lift from a friend) into Mastic Beach. I think the S71 might also work for certain parts of Mastic Beach.

LIRR leaves out of Penn Station.

If your hotel is across the street from Penn Station, simply cross the street. There are signs inside directing you to where the LIRR is (versus Amtrak and NJ Transit which also leave from Penn).

Trying to take a taxi for a distance less than a few blocks will likely get you laughed at.

To OP, in case you don't know. LIRR stands for Long Island Rail Road. There are several types of transportation inside Penn Station, so just be careful which signs you follow. You should buy your tickets ahead of time at the ticket windows inside Penn Station- that way you can ask for exact train times, directions, etc.

Don't buy tickets on the train itself- they charge an arm and leg in fees!

The Montuak line Services the Mastic-Shirley Station , Service is only during Rush hour from Penn Station. Offpeak you can take any train to Jamaica or Babylon and pick up a Montauk Line train bound from Mastic.
