There’s no street that can be argued to be the most iconic and/or important street in NYC that is also literally called Main Street. There are something like 5 streets named Main Street with one in each borough, I believe. None of them are the primary commercial or entertainment street of their boroughs, and certainly not for the entire city, though Main St/Roosevelt in Flushing in 2008 was, and probably still today is, the third busiest pedestrian crossing in the city.
As far as a most iconic/important street in NYC, whether or not it’s named Main Street, there have been topics on this and there are several options. However, the size of NYC makes it difficult to definitively select one street over all others for most people.

42nd is touristy for a couple blocks then turns into basic midtown office buildings (iow boring) plus NYPL and Bryant Park. I would consider Broadway a Main Street since it’s more of a nerve center that runs end to end the length of Manhattan.
That's what I thought when I read the OP. And it's also it's oldest street. It predates the start of the city of NY.

Manhattan is what most people think of when they think of NYC. It does function as the center of NYC. And at one time it was the entirety of NYC until the merger.
