When carrying a concealed weapon with a permit, is it legal to have a round in the chamber?

A round in the chamber?? As long as you have a permit to carry concealed, I don't see how they could tell you that you can't chamber a round. That would be insane and completely dangerous. I've had permits in three states(not NC) and they had no such restriction.
