I went to med school in Philly for 2 years and found a decent way back with only $7 in tolls. It takes an extra hour, but if you're not pressed for time (or madly in debt ), it's actually pretty good, and MUCH more scenic.
You take 22E to 81N then go through State College (322E, I believe) down to Harrisburg. At Harrisburg, you take 76E to Philly, which costs you ~$7.
Other notes:
>If this is your first time, I HIGHLY recommend a GPS! The exchange between 322E and 76E is confusing!
>The worst part is the first 30-40 miles on 22E. Lot of shopping centers, lots of slow drivers, traffic lights.
>You can bypass 76E by going down 30E through Lancaster, but this takes an extra 30-45 min.

Our kids live in State College, Harrisburg, and Philly. We take 22E to 90/322 to State College, then 322 all the way to Harrisburg. Then take I81 to I283 untill you reach 30E. follow 30 to 202N and that takes you to I76 and you can stay on I76 to Philly or get on the Turnpike for one or two exits to your destination. This is almost all 4 lane limited access except thru
Ebensburg and Blairsville. The 30 miles from Lancaster to Coatsville is a pain but if you time it before or after rush hours it only takes about 40 minutes thru to next part of 30 that is expressway until 202. We do it four time a year and it only takes 5-1/2 hours if you don't stop. It is only 35 miles more than the turnpike. this is from Monroeville. The same trip costs almost $50.00 both ways on the Turnpike. Thats almost 2 tanks or gas, and the price is going up 7% every year till 2030.
