What bus route should I take from Sky Harbor to Downtown Phoenix? Is it the Red Line?

Yes, it's the
Red Line (http://www.valleymetro.org/bus/Bus_Routes/Red_Line.htm - broken link) that you want; other routes stop at the Airport but they would require a transfer to get you Downtown. If you are arriving at Terminal 3 or Terminal 4, make sure to board the bus on the south curb. Buses that stop on the north curb are headed in the opposite direction toward Tempe.

is there any form of transportation other than taxi from phx sky harbor airport to hertz on so elliot rd and if so what are prices etc??

Yes, but it would involve multiple transfers -- not much fun with luggage. Why rent from Hertz all the way down in Ahwatukee when Hertz is at the Airport itself?

I would say catch a super shuttle (the blue vans) should be about $10
