Im one of those weirdos that have never taken an Uber or Lyft. Rather than have my friend wake up at 3:45am to drive me, do you think I will find an Uber from N Raleigh to RDU at that time? Am I taking any chances that I wont be able to find someone? This would be Wednesday morning.
I'm one of those weirdos too.. I schedule a regular taxi when I need to go to RDU in the wee hours of the morning (or any time really). Never had a problem with one not showing up.

when does your flight depart? 3:45 seems really early, even for the first departures of the morning. I know that TSA says get there XX hours beforehand, depending on domestic or International, but that just seems like an inordinately long amount of time for the first departures. Don't RDU departures start at 6:00AM?
I've taken these early AM flights out of RDU in the past, but I don't make a habit of it. Each time that I was there in the wee hours, it was a breeze through the process.
As to the question, I agree with others that say pre-schedule your Uber/Lyft. This would be especially true if you lived in way-way north Raleigh where drivers are less likely to be milling about.
I'd schedule a regular Taxi. Who knows if your Uber driver will wake up with a mean hangover and decide to go back to sleep.
At least a Taxi company has backups, worth the extra few bucks.
That's not how it works. Just because you schedule an Uber, that doesn't mean that it gets assigned to a driver at the time it's scheduled. It goes to an available driver the morning of. Just like a taxi.

So in other words there's no difference than just calling an Uber that morning.
From Uber's website:
"Scheduling a ride in advance does not guarantee you'll be connected with a driver. In the rare case a driver cannot be found, you'll be notified at the end of your selected time window."
Agree with m378 on this one. I work with a couple of young people who work Uber/Lyft on the side. Since they have day jobs, they aren't trolling for fares at 4 a.m. You might get lucky and find one who is but this is one instance I can't imagine going with a ride share service over a conventional taxi.
I once took a red eye flight from Denver to RDU. Got there around 5 AM.........the airport was packed and Uber had surge pricing so it might be possible that there are a lot of people going towards the airport at that time.

Yes, there is a difference. They are aware that there will need to be a driver in the area at a particular time. That's different than getting a call that a driver is needed there now. It allows for planning ahead.
