Is a larger house such as a 2 story home more expensive to maintain than a smaller square foot single story house? Are utility bills generally higher in larger homes?
Yes, of course, the basic cost in actual (net) operating/maintenance dollars is higher - (as well as financing/investment, insurance, utilities, maintenance/upkeep costs, etc.).
However, if those with larger (assumed 'nicer' or more upscale) homes also have proportionately larger incomes, the 'relative' cost-to-income/assets ratio is about the same or less.
Unaffordable discomfort with the cost arises when those without the higher incomes/assets ... attempt to appear as though they can afford larger, more expensive cars, houses, etc. -- with equal comfort as neighbors who actually have relatively higher incomes/assets.

Two stories isn’t “larger” by default. Just to clarify.
Heat rises so in summer or just generally in Florida, if you have a two story, you will pay more than a single story to cool it, assuming you don’t have a second air conditioner for the top level. Energy efficient windows help also.
The opposite can be true in winter but not so much in my experience.
Two stories have less roof to replace.
Two stories may or may not have a larger yard to care for.
Uhmmmm, yeah. If one house is larger than another that usually means more plumbing, more heating, more cooling, more flooring, more walls, more space, more of the many things that need to be paid monthly (utilities) and more of the things that need to be replaced and/or maintained like replacing flooring, painting, etc.
