If you want to check out some history, Sutter's Mill would be a good stop.

At the far north end, and a little ways off the pavement, there is Malakoff Diggins, which is known as one largest hydraulic mine. The remnants of the town of Bloomfield is also part of the park. You can wander around and look at the cabins and other buildings, or hike through the park.

The Empire Mine State Park in Grass Valley is a nice place to wander around.

Empire Mine SHP

Placerville has the Gold Bug Mining Tours.

There are also tours of a mine in Amador.

Sutter Gold Mine

More in Amador Co.

At Dog Town, you can take a diversion east of the Hwy 49 and drive past some mines. Take Main St/New Chicago Turn right (south) on Bunker Hill Rd. You can continue on Bunker Hill Rd, or take Fremont Mine Rd, which is gravel, but suitable for cars. I recommend Fremont Mine Rd as it takes you closer to some of the mines. It will merge with Bunker Hill Rd to the south, and you will find yourself in Sutter Creek. It is kind of a neat backroad that will allow you to see a few things a lot of people miss in the Motherlode.

Further south in Amador Co:

The current route of Hwy 49 goes right through the Argonaut Mining grounds in Jackson. You can see the mine from the highway. This is the mine wherein 47 miners were trapped in a fire. A local author wrote a good book about the Argonaut, and the neighbouring mine, The Kennedy Mine. There are tours of the Kennedy Mine's grounds. I don't know about the mine itself. Part of the fame of the Kennedy Mine is the elaborate system of moving ore around the grounds. There is a small park where you can walk around the remaining tailing wheels. Jackson: Kennedy Tailing Wheels

There is much more to see south of Jackson. You didn't mention if you plan to travel the length of Hwy 49 into Mariposa Co. What are your plans?

If you visit the railroad on the weekends there are some great train rides at night with beer and pretzil and wine and cheese runs. Used to ride on New Years Eve runs, lots of fun.

The train runs past my father in laws home, great to hear the train everytime we call him.
