I'm all about food when I'm there (well, maybe drinking too...lol)...

Have breakfast at Dolores Park Cafe or Harvey's on Castro St.
Walk across the Golden Gate Bridge
Grab a beer and tapas at the Thirsty Bear Brewing Co on Howard.
Get dessert from Citizen Cake on Grove St.
Relax with a Yellow Fever and dinner at the Poleng Lounge on Fulton
Shoot pool @ Trax on Haight.
S'Mores pie @ The Buckeye Roadhouse in Mill Valley
Irish Coffee @ The Buena Vista

But you can't be in San Francisco and not do the obligatory:
Fisherman's Wharf (sea lions make it worth it...that's about it)
Night tour of Alcatraz (the photo ops of the city once the sun goes down is sweet).
Lombard St.
Mels Drive-In
Chinatown
Japanese Tea Garden and the Conservatory of Flowers @ GGP
Palace of Fine Arts
Coit Tower
Shopping around Union Square

I go to San Francisco alot so I want to do/see something I haven't done or seen before there. I have already done all the super touristy stuff before. I just want to do something new there since this will be my last visit for a long time.

Would argue most culturally relevant place in SF region is Stanford campus, esp the Engineering side of campus....the ever-renewing, intellectual birthplace of SV and a rather enjoyable, scenic setting in which to actually walk around, as opposed to the filth/odors/crowds of most of City of SF....

Giants baseball game. Top of the Mark. Diva's & Tranny Shack (Stud). Olympic Club for golf. Golden Gate Fields (horse racing) That should keep you busy. Ripley
