Even if the driver can tolerate 18-20 hours straight in the car (which I find extremely unsafe), I highly doubt the kids could.
Have you ever tried it? I didn't think so. When I was a kid my parents took me and my three siblings on 1000+ mile drive straight through trips every summer. As a parent, I've taken my wife and two kids on many as well. For one thing, the kids will sleep 75% of the time. In any event, it's neither your place nor my place to tell her what her kids could tolerate.
Can you drive to LaGuardia? Spirit Airlines is efficient and cheap. I'm sure you can get advance tickets for about a hundred each round trip if you fly on a Tuesday or Wednesday. I fly often from Orlando and West Palm to Atlantic City, sometimes for as little as $58. Join the $9 dollar club.
A few suggestions that may help the OP and save her some money.
1. Rent the car from an off airport location. This will save you a bit money as most airports have substantial airport fees.
2. Be careful if renting a SUV or Minivan. These are usually the vehicles that have mileage and/or geographic restrictions. Sedans often don't have these restrictions. This is based on my experience with Hertz, could be different with other companies. If you are concerned about cargo space, consider mailing some things ahead of you. A smaller vehicle should also save some money on gas.
3. If you do drive try to fill up in South Carolina. Our gas taxes are next to nothing in comparison to NC, and less than GA. It won't save you a ton of money but maybe $10-20 roundtrip depending on how well you can time the fill ups.

Nope--teens and pre-teens are not going to sleep 75% of the way on a 20 hour trip.
+ 4 carryon bags ~$250.
+ want to sit together fee $ 1-50 so lets say $25x8 $200
and it goes on and on.. Do the Math ...
Agree, Spirit is a bad choice in this situation.
US Airways out of PVD is consistently one of the cheapest flights between New England and Florida. You can gate check your carry on sized bag for free.
I'm not sure where you live, but I just looked on Enterprise Rent a Car's website and a location in Hartford, CT had unlimited mileage on their vehicles according to their site.

However did you read the fine print regarding the geographic area the car can be driven in?
