Good question, and one that isn't discussed enough by travel lovers who live in America. We have a lot of great places right within our borders. I haven't traveled the US (or anywhere) as much as I'd like to, but here are some places that come to mind:
New York City:The absolute center of the universe for art, live music and energy. Always something interesting to see and do, even at 2:00am on a Monday.
New Orleans:You know you're in New Orleans when you're in New Orleans! A great sense of place, friendly southern vibe, interesting architecture and good food and drink!
San Francisco:I live here, so maybe not fair. But I do feel like I'm on vacation when I take road trips every other weekend!
Utah:The whole state is chock full of natural beauty.
I moved to my favorite vacation place (New Mexico) so I'm looking for a replacement favorite. Maybe Chicago or New Orleans for cities. Maybe coastal North Carolina or the Gulf side of Florida for casual beach time. There are still a few places I need to check out.
For urban vacation, my favorite is Seattle because there's cool stuff to do in and around the city (whale watching, waterfront shops, vibrant downtown).
For outdoorish (non-camping), renting a cabin by a lake in Minnesota. It's very pleasant.
For outdoors backpacking, the Selway-Bitterroot wilderness in Idaho. Vast mountain wilderness untouched since the early 1900s. Pristine and pure.
