Overnight, London to Edinburgh. Overnight Moscow to St Petersburg. Both times in sleeping berth.

Now if we're talking public transportation including airplanes - that's another story.

20 hours overnight on a bus trip from Aachen in Germany to Barcelona. The bus stopped every 2 to 4 hours, so it was tolerable

We spent a full day on the train from Melbourne to Sydney. I don't recall the precise number of hours.

The flight from Tampa to Melbourne involved 23 hours off the ground. We made 5 stops going and 5 returning.

We really enjoyed that trip.

It depends on the mode of public transportation:

Train - From London to a seaside resort in Italy (when I was a child with my father) which included an overnight seated (without sleeping berths) train from Paris to Italy and being on a ferry between England & France.

Ship - Moving from Marseille, France to Israel on a 5 day boat trip (as a child with my family)

Bus - A full day trip from Quito to Guayaquil, Ecuador.

Airplane - From Tel Aviv, Israel to Melbourne via London, Bangkok and Sydney. I started out early on a Friday morning, had an overnight at a hotel (provided for by the airline) in London and finally arrived in Melbourne towards midnight on a Sunday night (local time).

OP, do you just mean "transportation"? To me, public transportation means the subway, local buses, commuter trains, maybe ferries.

OP is this like one continuous route without changing planes or trains? And if so does it include layover time? Does this include cruise ships and overnight trains that are both transportation and lodging?
