I'll choose place that I haven't been to yet, and can't really afford a ticket to. Assuming visas were not an issue, I'll go with:
Timbuktu, Mali

Since I've been to over 50 countries already, many of them more than once

I'd pick 3 on my to go to list -

Antartica on Nat Geo Ice cutter with scientists (very expensive trip)

New Zealand

Kenya/Tanzania during Serengeti migration

1. Beijing, China (for Forbidden City, Great Wall, Chinese Government Tour, etc.)
2. Tel Aviv, Israel (for various Holy Land sites)
3. Cusco, Peru (for Machu Picchu/Amazon Rainforest)
Edit: (originally had Nairobi in my #3 spot. It's still a close #4, but a tad behind)
