Cartagena and Santa Marta on the coast are very touristy. I've stayed at the Charleston in Cartagena in the old city and ...wow...
Hoteles Charleston

Have also stayed at all-inclusive resort in Santa Marta which was lovely as well. Private huts off the beach. Gorgeous, peaceful and inexpensive.

As far as 'tourist friendly' cities...well that's all of Colombia. You'll find some of the friendliest people on earth. Tourism is still very green due to the violence and bad PR that followed it. But it's a whole new Colombia then even 10 years ago.

Medellin and Bogota are very metropolitan cities. Safe, clean and with a ton of activities (museums, night life, arts, etc).

The coffe regions - Quindio - Caldas - Pereira have unbelievable beauty.
It's much more of a quaint area. Many of the coffee haciendas have been converted to B & Bs. Here's some pics

Anyhow, depending on your wallet and your spirit much of Colombia is well worth your time.
