Bobby first then Charley - I honestly cried when both died and again every time I watch it in reruns. Then Jo and Ellen who aren't even on the poll. I loved their characters- cry over their "last" scene too. I really loved the alternate reality with Bobby and Ellen married. For a show that's so strong on masculine characters - and I love the Winchesters - I seem to like the strong women because Jody is a favorite too.
