I think I could possibly could be happy for 5 years in a lot of cities that I wouldn't necessarily otherwise pick the overall region- such as Nashville, Miami, Denver, Cleveland (or Cincinnati - which is better?) St Louis etc... maybe even Dallas/Ft Worth but otherwise don't care for the South, Midwest, Texas, Florida, Mid-Atlantic, Desert Southwest except maybe for a visit. I love NYC but you really need to be young and/or rich to have a good lifestyle there and alas, I'm neither.
Nice to see my city well represented here. Though I'll be the first to admit it is still somewhat lacking in a few categories though improving.
Every one of these towns is VERY special in one way or another.
I would NEVER consider:
Detroit
Cleveland
Buffalo
Rochester (MN or NY)
NYC
Mostly because their weather sucks majorly.
That being said, the "never" list has some magnificent suburbs and I would consider living in any of those, as long as I could out four months a year to a decent climate.

Yep! Neighborhoods DO count, and being able to escape the worst weather months (also a big reason for my preference) is a huge thing. Most every city that I can think of would only have at most 20% of areas or neighborhoods that I would ever consider living in, my own included, and those areas would generally be expensive (basically, first ring street car suburbs just on the edge of downtown, that are safe, green, progressive and full of great historic housing stock) to live in and limited in area. If I can find that in most cities I'm gonna find just about anything tolerable and for many cities it would suit me just fine regardless of the surrounding area.
