What can I do or say to not work weekends or nights? Is there a good excuse for this?

Very few vets are open on Sunday. Most are open at least half a day on Saturday. It wouldn't surprise me if they asked you to work on some Saturdays. If you are unwilling to work on the weekend, I think it will limit the places that are willing to hire you. I'd ask them about weekends when you interview and see if their schedule will fit yours. I think each vet's office will be different.
What kind of job? If it is a restaurant or retail, or some business that is open at night and on weekends, don't plan on getting hired if you are unwilling. If it is truly occasionally, then suck it up, and do it when needed.
