They spider all the job sites and it goes on to their site. Very handy. Craigslist for local jobs but mostly used by smaller companies. I get spam all the time from using Monster. Slow site which I never liked.
Actually try and network and get into industry specific unemployment groups. Make sure you join linkedin.com if you are a white collar professional.

It depends on the job you're looking for. As mentioned above, LinkedIn is a professional networking community and a number of the jobs listed there state that they are LinkedIn exclusives. I have an interview with one of those this week.
Cybercoders is another decent one for technology professionals
Marketingjobs. com is rather obvious
Careerbuilder is decent- mostly because most of the postings list the hiring company
Monster.com is a mess and results in more spam than leads
Indeed.com is a decent consolidator
6figurejobs.com requires a fee
Marketing-jobs.theladders.com also charges a fee
If you're receiving UI, your state's unemployment site should also have a job board
There are many others, so focus on your industry or type of employment. But remember, only 10% of white collar jobs are filled through these job boards. Your best option is to find the hiring company and go direct.

Agree with JTGJR - you're best option is always go direct. Although some only advertise through these sites. When I looked, I used all of them, and when I found a job I liked - I actually went to that company's website to see if it was posted there. That was always my first avenue with online job applications.
Of course, depending on the position, networking is probably the most powerful tool. There's nothing like having a mutual friend/colleague recommend you.

I like Americas Job Exchange, Indeed, and Simply Hired. Direct contact is best, but you can get the leads from these sites and many others.

Here's a first hand example. About a month ago I spotted a good position at a company website. Against my better judgment, and because I wanted to be able to report some activity for my UI, I applied through the company website.
As expected, nothing happened. Last week, I noticed the position was still open, so I did some LinkedIn research and found a person at the company with whom I had worked 10-years ago. I connected with him through LinkedIn, and he provided me the direct email to the internal company recruiter. I emailed my cover and resume directly to her and she called me within 30-mins. I had a phone interview with the hiring manager within 24-hrs and received a callback a few days later for a face-to-face.
I'm the same person and I submitted the same resume but got different results. Blind submission- nothing. Network support- opportunity.
