The main hub, gathering place, with a lot of neon billboards, jostling crowds, the main focus for the city with an emphasis on commercial uses. Where all the action is. Not necessarily the official 'centre of town' if there is such a thing.
In London I would guess Piccadilly Circus, in Tokyo maybe somewhere in Shinjuku?
In São Paulo, the most similar place would be the Paulista Avenue in terms of being an outstanding hub for business and services, and it's also the most visited address in the city, but still very different from Times Square, because the law forbids billboards, neons and totems. Aesthetically it has nothing special, it looks like any boulevard with high rises in the world.
Chicago it might be either the Magnficent Mile (Michigan Avenue) or Millennium Park (where the reflecting bean is) in relation to where all the tourists go. It's not where "all the action is" though, just one of them. Any world class city is going to have many areas of "action" in it.

I would suggest it could be argued that in London the unofficial 'centre' is not Piccadilly Circus but Trafalgar Square.
