I have never been to Australia and might be coming to visit the Melanoma Institute/Poshe Center, which I believe located
40 Rocklands Road, North Sydney, NSW 2060, lower North Sydney.

Can anyone help me with hotel suggestions and the best place to stay (not too expensive). I didn't know if it was best to stay near that area or to stay in Darling Harbour area and travel to the institute.

Any help would be greatly appreciated!! Thanks

Most NSW ""out of towners"" that head into Sydney tend to stay in North Sydney, as it is still close to the city, Luna Park, Taronga Zoo etc and other attractions.
North Sydney has plenty of available accomodation, usually a lot cheaper than staying in the city itself.
This is a popular one:

North Sydney Harbourview Hotel, Hotel and Accommodation in North Sydney, Sydney

Otherwise have a look at:

www.wotif.com.au

www.lastminute.com.au

To name just a few.
Rydges North Sydney is also ok, just google the link.

The only one I can think of is Vibe Hotel -- not sure if it is expensive or not... but it is in Milsons Point, very close to Nth Sydney

I don't think staying in the Darling Harbour area would be a bad option, North Sydney is only 2 stops on the train across the Harbour Bridge and you can enjoy all that the city has to offer. Sydney Hotels, though, are quite unreasonably priced. Get an apartment to share.

There's a new YHA at the Rocks which might be good.
