It does not, however, have the ability to give directions and time/distance to the on ramp of a highway. For ex, if I want to know how long it's gong to take to drive from the house at 17 Main St. Anywhere, USA to the onramp of the nearest hwy, MQuest does not do that. At least, not as far as I can tell.
You can use Google Maps to plot a trip using GPS-coordinates.
Just pull-up the regular map view on Google Maps (Layers - terrain). Then zoom to the area you want to plot as your starting-point, double-click that area and it will pop-up a GPS-coordinate, click on it and it will go into the trip-plotting dialogue-box. Double-click on your destination and it will again pop-up GPS-coordinates...copy and paste in the trip-plotter as your destination.
What exactly are you trying to do? Are you trying to drop a pin on the nearest freeway entrance and then get directions to it? Or are you expecting MapQuest to find the nearest freeway entrance on its own, then route you to it?

For what it's worth, there is an easier way in Google Maps - if you ask for directions between any two locations, specified by their addresses only, and then pick the desired route of several proposed alternatives, then click the "Details" link in the text describing the route - it will show you the distances along segments of the route between intersections where you change roads - so you can see how many miles you are traveling along each road, as well as the estimated travel times for each segment.
Just google RT ?"I really should know how to get here but I don't" from current location and then look at the map that comes up and you'll see a blue or grey line. move the map around to look at what seems to be the most straight forward route and take a snapshot in your head. At the point where you lose familiarity note right or left and name of street (write on hand or large letters quickly glanceable on a sheet of paper), start car and go.
If new to the area or new to driving, drive around a bit for a few days after getting settled to learn the quickest and back-up routs to go out of town.

Anyone know of a site that does this?
As far as I know all directional software used for driving should do that. But you have to enter a destination point and your location. For example, in your case your location is your address at "17 Main Street," so when you enter the destination point, lest say, "Highway 95", the software creates a route for you to travel, and the time it takes to get there. Google Maps, Apple Maps, and so on do that for you. If you have an iPhone, it already has the app "Maps."
If you drop a pin anywhere on Google maps you can get directions to it from any starting point. It’s as easy as finding the location on the map. Holding your finger on it until it drops a pin, then put in your starting point, and it’ll give you directions and time for if you want to drive or walk there. Doesn’t get much easier than that…
