What to do? Walk around, hike, relax, absorb the beauty and power there ... just Be there.
I'd wait until Tioga pass is open (late May or June) so that you can drive up into the high country of Yosemite.
June is a beautiful time to go. The water is really flowing and if you can be there when the moon is full, that would be great. You may even see a moon-bow.

See some of the waterfalls (Briedelveil, etc), and the view of El Capital and Half Dome after you drive through the tunnel (depending though on which way you enter Yosemite). I can't think of the name of it, but it's like a stop on the main road that goes to the village, and it's right after you drive through a tunnel. Go to The Awahnee Hotel. Take lots of pictures, walk around, go to the village and buy a souvenir. Try to go on a weekday or off-season when there is less people and it's more enjoyable. It can get VERY crowded in the summer. Although I guess it is also pretty then. If you are staying for awhile, maybe stay at Curry Village.

I love to eat fresh trout but haven't a clue how to bait a hook. I fished once with my dad when i was a child and the only memory i have is of him whacking a live fish over a big rock. Bleccccccchhhhhhhhhhh Why did he do that?!
But another thing comes to mind regarding Yosemite. If it's warm enough when you go and you like cold river water (which i love) find a turn off by the Merced river and search for a good swimming hole. There are some great ones and the water is divine!
